Humanigen, Inc. is a clinical-stage biopharmaceutical firm primarily focused on addressing and mitigating an exacerbated immune response termed "cytokine storm." At the forefront of its therapeutic efforts is lenzilumab, an antibody engineered to bind with and neutralize granulocyte-macrophage colony-stimulating factor (GM-CSF). The company is heavily invested in advancing lenzilumab as a treatment for cytokine storm associated with COVID-19; it has successfully completed a crucial Phase 3 study and is concurrently participating in an NIH-sponsored Phase 2/3 investigation for this indication. Beyond COVID-19, lenzilumab's utility is being explored in combination with CD19-targeted CAR-T cell therapies. Ifabotuzumab, which targets EphA3, completed a Phase 1 study for glioblastoma multiforme, demonstrating precise targeting of all known tumor locations without affecting healthy tissues. The second, HGEN005, designed to target EMR1, is being investigated for various eosinophilic conditions, including eosinophilic leukemia, both as an optimized standalone antibody and as a foundational element for a new CAR-T therapy.
